Ponceau S Staining Solution is made using Ponceau S. Ponceau S is a sodium salt used for the detection of proteins on PVDF, and nitrocellulose membranes. Staining of blot transfer membranes permits visualization of proteins and allows the extent of transfer to be monitored. Ponceau S binds rapidly and reversibly to the positively charged amino groups and binds non-covalently to non-polar regions in the protein. Ponceau S stain is designed for rapid (5 min) staining on nitrocellulose or PVDF membranes and also for staining protein on cellulose acetate membranes. Microgram quantities of transferred protein will be detected as red protein bands with a clear background. Ponceau S staining on nitrocellulose and PVDF is reversible by washing the stained membrane with dH2O or blocking buffer, this allows further immunological detection.
Store at room temperature. The product is at working concentration, no further dilution is necessary. The limit of detection for this stain is 200 nanograms of protein after separation by electrophoresis in polyacrylamide gels and transferred to nitrocellulose or PVDF membranes. Ponceau S is not suitable for use with nylon membranes.
